Free Agent gave The Queen a first winner at the big fixture since 1999 with a battling success in the Chesham Stakes.
Held up in the rear by Richard Hughes in the early stages, the Richard Hannon-trained 7-2 joint-favourite came from last to first to win by two and a quarter lengths from Seaway.
Her Majesty received rapturous applause when collecting her trophy from former champion jump jockey Richard Dunwoody, and she said to Hughes: "It is very exciting."
"That is what racing is all about - you never know what is going to happen."
Hannon said: "I think he will make into a really nice horse and would be more of a Derby prospect than a Guineas horse."
Macarthur gave trainer Aidan O'Brien and jockey Johnny Murtagh their fifth win of the week with victory in the Hardwicke Stakes, while Honolulu swiftly made it six of the best for the deadly duo in the concluding Queen Alexandra Stakes.
Linda Perratt's Big Timer struck a blow for Scotland with a 20-1 success in the Wokingham Stakes, while Sugar Ray made virtually all the running in the hands of Ryan Moore to win the Duke of Edinburgh Handicap.
